Claudins are tetraspan proteins making the tight junction. Claudins have four transmembrane domains, two extracellular loops (ECLs: ECL1 and ECL2), amino- and carboxyl-terminal cytoplasmic domains, and a short cytoplasmic motif.
Claudin-5 is primarily expressed by the endothelium in the brain and the retina, making the blood-brain barrier and the blood-retina barrier respectively.
