Cas9 mRNA is a capped and polyadenylated messenger RNA encoding Cas9 protein fused to a nuclear localization sequence (NLS).

CRISPR Associated Protein 9 (Cas9) functions as part of the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) genome editing system. In the CRISPR system, an RNA guide sequence targets the site of interest and the Cas9 protein performs double stranded DNA cleavage.

The Cas9 mRNA expresses the version of the Streptococcus pyogenes SF370 Cas9 protein. The Cas9 mRNA is capped using m7G(5′)ppp(5′)A analog and polyadenylated to mimic a fully processed mature mRNA. The Cas9 mRNA can be used in pronuclear injection to mouse or rat embryo to create knockout or knockin transgenic animals.
