Claudin-18 is a protein that is encoded by the CLDN18 gene in human. Claudin-18 belongs to the Claudin protein superfamily.
Claudins are tetraspan proteins making the tight junction. Claudins have four transmembrane domains, two extracellular loops (ECLs: ECL1 and ECL2), amino- and carboxyl-terminal cytoplasmic domains, and a short cytoplasmic motif.
Claudin-18 has two splicing isoforms: 18.a1 and 18.a2. The difference is within the ECL1 domain. CLDN18.a1 is expressed in the lungs and CLDN18.a2 is expressed in the stomach. CLDN18.a2 is an important marker for gastric cancer and pancreatic cancer.
This antibody specifically recognizes Claudin-18.a2 isoform. It shows no affinity to Claudin-18.a1 isoform.
