Discs large homolog (DLG1), also known as synapse-associated protein 97 (SAP97), is encoded by the dlg1 gene in human. Mammalian DLG1 protein is homologous to Drosophila protein Dlg.
DLG1 is a cytosolic scaffold protein consisting of three PDZ domains, an SH3 domain, a I3 domain and a MAGUK domain. Drosophila Dlg1 protein plays a vital role in establishing the apicobasal polarity. Mammalian DLG1 protein is expected to perform a similar function. In addition, mammalian DLG family proteins have been shown to regulate adherens junction assembly, signal transduction, cell proliferation, synaptogenesis and lymphocyte activation.
