Large tumor suppressor kinase 2 (LATS2), also known as KPM, is encoded by the LATS2 gene in human. LATS2 is a negative regulator of p53 and may function in a positive feedback loop with p53 that responds to cytoskeletal damage. LATS2 is part of the Hippo signaling pathway. The Hippo signaling pathway comprises of a kinase cascade wherein MST1/2 phosphorylates and activates LATS1/2, which in turn phosphorylates but inactivates YAP and WWTR1/TAZ. Phosphorylation of YAP1 by LATS1/2 inhibits its translocation into the nucleus to regulate cellular genes important for cell proliferation, cell death, and cell migration.
