Plakophilin-1 is encoded by the PKP1 gene in human. Plakophilin-1 belongs to the protein family of Plakophilin. Plakophilin proteins contain nine armadillo repeats, localize to the desmosomes, and participate in linking cadherins to intermediate filaments in the cytoskeleton.
Plakophilin-1 has been shown to interact with Desmoplakin and regulate epidermal morphogenesis. Mutations in Plakophilin-1 cause autosomal recessive ectodermal dysplasia and skin fragility syndrome.
