Smoothened (SMO), also known as frizzled class receptor 11 (FZD11), is encoded by the SMO gene in human. Smoothened is a key transmembrane protein participating in the hedgehog signaling pathway.
Smoothened consists in seven transmembrane domains, a N-terminal extracellular domain and a C-terminal intracellular domain. Smoothened interacts with the patched protein, a receptor for hedgehog ligand proteins. Binding of sonic hedgehog (SHH) to its receptor patched is thought to prevent normal inhibition by patched of smoothened.
