The Na-Cl cotransporter (NCC) transport Na+ and Cl- at the molecular ratio of 1:1. It belongs to the SLC12 cotransporter family and is known as SLC12a3. In human, it is encoded by the SLC12A3 gene.
NCC consists in 12 transmembrane domains and is highly expressed in the kidney. Mutations in NCC cause Gitelman syndrome (OMIM 263800), which features low blood pressure, hypokalemic metabolic alkalosis, hypomagnesemia and hypocalciuria.
