Roundabout homolog 3 is a membrane protein that is encoded by the ROBO3 gene in human. Human ROBO3 protein is structurally similar to a Drosophila integral membrane protein that is encoded by the Drosophila roundabout gene (a member of the immunoglobulin gene superfamily). ROBO3 protein regulates axonal navigation at the ventral midline of the neural tube.
Mutations in ROBO3 gene result in horizontal gaze palsy with progressive scoliosis (HGPPS); an autosomal recessive disorder characterized by congenital absence of horizontal gaze, progressive scoliosis, and failure of the corticospinal and somatosensory axon tracts to cross the midline in the medulla.
