P21Cip1 (alternatively p21Waf1), also known as cyclin-dependent kinase inhibitor 1 or CDK-interacting protein 1, is a cyclin-dependent kinase inhibitor that is encoded by the CDKN1A gene in human. The P21 protein binds to and inhibits the activity of cyclin-CDK2, -CDK1, and -CDK4/6 complexes, and thus functions as a regulator of cell cycle progression at G1 and S phase. The expression of the P21 protein is tightly controlled by the tumor suppressor protein p53, through which P21 mediates the p53-dependent cell cycle G1 phase arrest in response to a variety of stress stimuli.
