Nephrin is encoded by the Nphs1 gene in human. Nephrin is a structural protein making the slit diaphragm in the glomerular podocytes. Slit diaphragm is a specialized cell junction that constitutes the glomerular filtration barrier in the kidney.
Nephrin consists of an extracellular N-terminal domain, a helical transmembrane domain, and a cytoplasmic C-terminal domain. The N-terminal domain in nephrin, resembling that in classic cadherin, mediates trans-interactions between podocyte foot processes.
Nephrin is exclusively expressed by the glomerular podocytes in the kidney. Mutations in nephrin cause congenital nephrotic syndrome of the Finnish type (OMIM 602716).
