Desmoglein-3, also known as Cadherin family member 6 (CDHF6), is encoded by the DSG3 gene in human. Desmoglein-3 is a member of the desmoglein protein subfamily. Desmogleins, along with desmocollins, are cadherin-like transmembrane glycoproteins that are major components of the desmosome.
Desmoglein-3 contains an amino-terminal extracellular domain or ectodomain or EC domain that is followed by a transmembrane domain and a carboxyl-terminal intracellular domain. The extracellular domain mediates intercellular interactions in the desmosome whereas the intracellular domain mediates Desmoglein-3 interaction with Plakophilins.
Desmoglein-3 is highly expressed in the skin epithelium as well as in the cardiac muscle. In the skin, Desmoglein-3 is expressed in the basal layer of the epidermis. In the cardiac muscle, Desmoglein-3 is localized to the intercalated discs which mechanically couple cardiac cells to function in a coordinated syncytial structure.
