Protocadherin FAT4 is a protein that is encoded by the FAT4 gene in human. FAT4 is also known as FAT-J or CDHF14. This gene is an orthologue of the Drosophila fat gene. FAT is involved in establishing the planar polarity in Drosophila cells.
Like classic cadherins, protocadherin proteins contain an amino-terminal extracellular domain or ectodomain or EC domain that is followed by a transmembrane domain and a carboxyl-terminal intracellular domain. Compared to classic cadherins, the ectodomains in protocadherins are much larger.
FAT4 plays a role in regulating planar cell polarity (PCP). Mutations in this gene have been associated with Van Maldergem Syndrome 2 (OMIM 615546).
