DNA methylation is the major modification of eukaryotic genomes and plays an essential role in mammalian development. Methyl-CpG-binding domain protein 4 is a DNA glycosylase that is encoded by the MBD4 gene in human. MBD4 contains an MBD domain at the N-terminus that functions both in binding to methylated DNA and in protein interactions, and a C-terminal mismatch-specific glycosylase domain that is involved in DNA repair.
