BiCell Scientific Inc’s Mito-binder is a type of fluorophore based on carbocyanine that accumulates in active mitochondria but is not well-retained in mitochondria of pre-fixed cells. The Mito-binder can freely diffuse across the plasma membrane and the mitochondrial membrane. Because Mito-binder contains a thiol-reactive chloromethyl group, mitochondrial staining can be retained after the mitochondrial membrane potential is lost during cell fixation. Therefore, Mito-binder is suitable for labeling live cells as well as post-fixed cells but not suitable for labeling histological tissue section.

Staining of cell culture
- Dilute Mito-binder Fluorophore Conjugate at 1:100 in PBS or cell culture medium.
- Add diluted Mito-binder to cell culture and incubate for 1hr at 37oC.
- Aspirate cell culture medium and fix cells in 3.7% PFA for 1hr at 4oC.
- Image labeled cells with fluorescent microscopy.
