Podocin is encoded by the Nphs2 gene in human. Podocin is a structural protein making the slit diaphragm in the glomerular podocytes. Slit diaphragm is a specialized cell junction that constitutes the glomerular filtration barrier in the kidney.
Podocin has a transmembrane domain forming a hairpin structure, with two cytoplasmic ends at the N- and C-terminus, the latter of which can interact with nephrin.
Podocin is exclusively expressed by the glomerular podocytes in the kidney. Mutations in podocin cause steroid resistant nephrotic syndrome (OMIM 604766).
